It proved, once and for all, that she wasn’t a jealous bitch (though much of the fandom continues to think otherwise…). She had watched Sasuke descend with her sixth sense and witnessed his fall with her own body - his stabbing her was his final “sacrifice,” as it was referred to in the chapter. And she knew Sakura loved him too, but she didn’t want to see her die.
2. MY CREYS
No one can witness this and in good faith say Karin’s love for Sasuke was superficial. We don’t know the exact details as to why it happened, but we know that her entire family and village were destroyed (maybe because she’s an Uzumaki? I think Kishi will explain this sooner or later). She then went to Orochimaru because the only soul left on earth with whom she was acquainted was Uchiha Sasuke, the boy who had saved her. And this is the root of her love for him - to see him smile again like he did, purely and sincerely on that day long ago. She wants, even above receiving his love in return, to see him happy. And when she sees that he has crossed the moral event horizon and she cannot help him, she leaves him, even if she still loves him. I have so much respect for her for that.
3. When Sakura heals her, Karin cannot help but empathize with her. It is significant that when she cries with Sakura over their shared love for a man who is not what he once was, this is only the second time we have ever seen her cry. The other was when she shed a single tear because she thought the Tsuchikage had obliterated Sasuke.
The rest of what she says is “Don’t cry like that in front of me…!” Then, against her will, she, too, begins to weep.
4. When she drags a weakened Sasuke and Juugo out of the way of the Hachibi, almost sacrificing her own life in the process. (She is set on fire by the Hachibi in the panels that follow.)
5. When Sasuke tells Karin she saved him. He says this to her, specifically, and says nothing of the sort to Suigetsu and Juugo. The way Kishi singles out his relationship with her (even drawing the two of them close to each other in typical scenes - like when Taka is running and jumping through one of those huge-ass Naruto forests) tugs at my heartstrings. He doesn’t just say thank you, he acknowledges what she’s done for him. And she just replies with a simple “Aa” which is like saying “yeah,” in Japanese. It’s so touching. They sit in a heap afterwards, panting, recovering from their wounds.
6. Then, there is the fight with Danzo. Again, she risks life and limb for Sasuke when he is unable to move because of Danzo’s binding curse, doing so to the point of being illogical, as she stood no chance whatsoever against Danzo, and she knows it. So does he, as you can see in the panels below. He kicks her away instead of attacking her physically, because he is trying to conserve his chakra, as Tobi observes.
According to the Databook, Karin is from a village with few to no shinobi that was destroyed when she was 12, which explains not only her physical weakness, but also the reason why she does not run like a shinobi, with her arms stiffly at her sides, and why she knows few jutsu. She works at a disadvantage, using her combination of keen intellect, sensing, and healing abilities.
7. One thing that’s interesting about Karin is that her love for Sasuke has never gotten in the way of her getting the job done. Perhaps one of the best examples of this is the following page from the manga.
Notice that she is relieved - but she does not voice that thought aloud. Instead, she is all business. She tries to get him back into battle.
8. The Databook specifically says that “Not only is she able to use sensory skills, but Karin has extensive knowledge at her disposal. With this, she comes up with brilliant strategies that contribute to the mission.” An example of this is when she throws Konoha’s tracking dogs off Taka’s scent. And with regard to her sensory powers, Kishi says that they “surpass ocular jutsu.” [X]
Even Kakashi points out how good her plan was for scattering Sasuke’s scent by tearing up his old shirt and having Juugo’s birds scatter the pieces to throw Konoha’s tracking dogs off the scent:
9. In the animé, when Karin is bitten, Karin vision is all shoujo and sparkly. Yet in the manga, the whole process is overtly sexual and isn’t played for laughs at all. This is a pretty big deal in a shounen manga, in which characters are rarely physically intimate even if fanservice is frequent. (And this is actually another reason why this ship stands a chance - Kishi basically had them having sex in public, and unlike the animé, he took it seriously.) Kishi never once cracked a joke about this, and in fact, Sasuke and Karin were the only ones to discuss the whole biting process at all. I imagine that most of her scars are from Sasuke, but it is doubtful that all of them are, given that this ability is entirely the result of Kabuto and Orochimaru’s experimentation, so she was probably bitten by other people as well. Any bites from someone other than Sasuke were probably equivalent to rape (since I doubt she wanted some rando, or worse yet, Orochimaru or Kabuto, biting her), making the ability and the themes it introduces dark indeed for a “children’s” manga.

It is interesting that because of this Karin is the only person with whom Sasuke has been so physically intimate. She literally gave of herself to him, shared her chakra with him, her essence.
10. This moment exhibits how human Karin is in spite of her tough exterior, and how deep her love for Sasuke is.
In the manga, she has only smiled once (all the other times were smirks). This, of course, was one of the two times she cried as well.
